Tadalafil has a half-life of 17.5 hours and its effects can last up to 36 hours in some patients. How Tadalafil Works

Tadalafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5). Tadalafil has the chemical structure of a pyrazolo[4,3-d]pyrimidine. Its empirical formula is C22H19N3O4, and its molecular weight is 389.41. The structural formula is: Tadalafil inhibits PDE5. PDE5 is an enzyme found in certain body tissues, including the penis. Inhibiting PDE5 increases blood flow to the penis. Tadalafil was approved by the United States (U.S.) Food and Drug Administration (FDA) in 2003 for the treatment of erectile dysfunction (ED). ED is the inability to get or maintain an erection. Tadalafil is also used to treat pulmonary arterial hypertension and benign prostatic hyperplasia, two conditions that can cause ED. In clinical trials, tadalafil was shown to be effective in treating ED in men of all ages and races. The most common side effects of tadalafil are headache, indigestion, back pain, muscle aches, flushing, and stuffy or runny nose. These side effects usually go away after a few hours. Men who use tadalafil may have an increased risk of developing skin cancer. 

Tadalafil Dosage

Tadalafil is a PDE5 inhibitor indicated for the treatment of erectile dysfunction (ED). The recommended starting dose of tadalafil for use as needed for most patients is 10 mg taken orally approximately one hour before sexual activity. Rare but serious side effects associated with tadalafil use include: Vision changes or sudden vision loss, Ringing in the ears or sudden hearing loss, Severe dizziness or lightheadedness, Priapism (prolonged erection lasting more than 4 hours). 

Drug Interactions with Tadalafil

Tadalafil is a prescription medication used to treat erectile dysfunction (ED). It works by relaxing the muscles and increasing blood flow to the penis. Tadalafil can be taken with or without food. Common side effects of tadalafil include headache, flushing, upset stomach, stuffy nose, and muscle aches. Tadalafil can also cause dizziness. Tadalafil may interact with other medications that you are taking. It is important to tell the medical professionals about all medications that patient's are taking, including over-the-counter drugs, vitamins, and herbs. Patient's should not start or stop taking any medication without permission of doctor. Some medications that may interact with tadalafil include: Alpha blockers such as alfuzosin (Uroxatral), doxazosin (Cardura), prazosin (Minipress), silodosin (Rapaflo), tamsulosin (Flomax), Azole antifungals such as itraconazole (Sporanox) or ketoconazole (Nizoral), Boceprevir (Victrelis) or telaprevir (Incivo) for hepatitis C infection, Certain antibiotics such as clarithromycin (Biaxin).
