Abstract

As drug therapy is prolonged, a decline in receptor responsiveness can undermine therapeutic efficacy, complicate disease management, and contribute to adverse outcomes. Functional assays, such as insulin sensitivity tests or dynamic endocrine testing, can support the diagnosis. In research settings, receptor density and signaling may be assessed using radioligand binding or molecular imaging technologies, though these are not routinely available in clinical practice.

Treatment & Management

Management strategies for receptor desensitization focus on restoring drug responsiveness and optimizing therapeutic outcomes. Approaches include drug holidays or intermittent dosing protocols to allow receptor resensitization, switching to alternative agents with different mechanisms of action, and combination therapy to target multiple pathways. Dose adjustment should be guided by careful monitoring of clinical and laboratory parameters.
