Parkinson's disease (PD) is a progressive neurodegenerative disorder that primarily affects dopaminergic neurons in the substantia nigra. It is characterized by motor symptoms such as tremors, rigidity, and bradykinesia, as well as non-motor symptoms including cognitive impairment and mood disorders. This article aims to provide a comprehensive overview of PD for healthcare professionals.

Etiology and Pathophysiology

The exact cause of PD remains unknown, but it is thought to involve a combination of genetic and environmental factors. Pathologically, PD is characterized by the loss of dopamine-producing neurons in the substantia nigra and the presence of Lewy bodies, which are protein aggregates primarily composed of alpha-synuclein. The disease progression is typically asymmetric, with symptoms appearing on one side of the body before the other.

Diagnosis and Management

PD is primarily diagnosed based on clinical criteria, with neuroimaging used to rule out other causes of parkinsonism. The management of PD involves a multidisciplinary approach, with pharmacological treatment aimed at managing symptoms and improving quality of life. Levodopa remains the most effective medication for motor symptoms, while non-pharmacological interventions include physical and occupational therapy.
